ABSTRACT

OBJECTIVE@#To observe the effect of Tongdu Tiaoshen (promoting the circulation of the governor vessel and regulating the spirit) electroacupuncture (EA) pretreatment on pyroptosis mediated by peroxisome proliferators-activated receptor γ (PPARγ) of the cerebral cortex in rats with cerebral ischemia reperfusion injury (CIRI) and explore the potential mechanism of EA for the prevention and treatment of CIRI.@*METHODS@#A total of 110 clean-grade male SD rats were randomly divided into a sham-operation group, a model group, an EA group, an EA + inhibitor group and an agonist group, 22 rats in each group. In the EA group, before modeling, EA was applied to "Baihui" (GV 20), "Fengfu" (GV 16) and "Dazhui" (GV 14), with disperse-dense wave, 2 Hz/5 Hz in frequency, 1 to 2 mA in intensity, lasting 20 min; once a day, consecutively for 7 days. On the base of the intervention as the EA group, on the day 7, the intraperitoneal injection with the PPARγ inhibitor, GW9662 (10 mg/kg) was delivered in the EA + inhibitor group. In the agonist group, on the day 7, the PPARγ agonist, pioglitazone hydrochloride (10 mg/kg) was injected intraperitoneally. At the end of intervention, except the sham-operation group, the modified thread embolization method was adopted to establish the right CIRI model in the rats of the other groups. Using the score of the modified neurological severity score (mNSS), the neurological defect condition of rats was evaluated. TTC staining was adopted to detect the relative cerebral infarction volume of rat, TUNEL staining was used to detect apoptosis of cerebral cortical nerve cells and the transmission electron microscope was used to observe pyroptosis of cerebral cortical neural cells. The positive expression of PPARγ and nucleotide-binding to oligomerization domain-like receptor protein 3 (NLRP3) in the cerebral cortex was detected with the immunofluorescence staining. The protein expression of PPARγ, NLRP3, cysteinyl aspartate specific protease-1 (caspase-1), gasdermin D (GSDMD) and GSDMD-N terminal (GSDMD-N) in the cerebral cortex was detected with Western blot. Using the quantitative real-time fluorescence-PCR, the mRNA expression of PPARγ, NLRP3, caspase-1 and GSDMD of the cerebral cortex was detected. The contents of interleukin (IL)-1β and IL-18 in the cerebral cortex of rats were determined by ELISA.@*RESULTS@#Compared with the sham-operation group, the mNSS, the relative cerebral infarction volume and the TUNEL positive cells rate were increased (P<0.01), pyroptosis was severe, the protein and mRNA expression levels of PPARγ, NLRP3, caspase-1 and GSDMD were elevated (P<0.01); and the protein expression of GSDMD-N and contents of IL-1β and IL-18 were increased (P<0.01) in the model group. When compared with the model group, the mNSS, the relative cerebral infarction volume and the TUNEL positive cells rate were decreased (P<0.01), pyroptosis was alleviated, the protein and mRNA expression levels of PPARγ were increased (P<0.01), the protein and mRNA expression levels of NLRP3, caspase-1 and GSDMD were decreased (P<0.01), the protein expression of GSDMD-N was reduced (P<0.01); and the contents of IL-1β and IL-18 were lower (P<0.01) in the EA group and the agonist group; while, in the EA + inhibitor group, the protein expression of PPARγ was increased (P<0.01), the protein and mRNA expression levels of NLRP3 and GSDMD were decreased (P<0.01, P<0.05), the mRNA expression of caspase-1 was reduced (P<0.01); and the contents of IL-1β and IL-18 were lower (P<0.01). When compared with the EA + inhibitor group, the mNSS, the relative cerebral infarction volume and the TUNEL positive cells rate were decreased (P<0.05, P<0.01), pyroptosis was alleviated, the protein and mRNA expression levels of PPARγ were increased (P<0.01), the protein and mRNA expression levels of NLRP3, caspase-1 and GSDMD were decreased (P<0.01), the protein expression of GSDMD-N was reduced (P<0.01); and the contents of IL-1β and IL-18 were declined (P<0.01) in the EA group. Compared with the agonist group, in the EA group, the relative cerebral infarction volume and the TUNEL positive cells rate were increased (P<0.05, P<0.01), the mRNA expression of PPARγ was decreased (P<0.01) and the protein expression of GSDMD-N was elevated (P<0.05); and the contents of IL-1β and IL-18 were higher (P<0.01).@*CONCLUSION@#Tongdu Tiaoshen EA pretreatment can attenuate the neurological impairment in the rats with CIRI, and the underlying mechanism is related to the up-regulation of PPARγ inducing the inhibition of NLRP3 in the cerebral cortex of rats so that pyroptosis is affected.

ABSTRACT

Depression is a common emotional disorder that seriously affects people's life and health all over the world. The pathogenesis of depression is complex, and traditional Chinese medicine (TCM) for antidepressants has a good therapeutic effect because of its multi-component, multi-pathway, and multi-target action mode. At present, the anti-depressive mechanism of TCM has not been fully clarified, but it is clear that depression is closely related to metabolic health. Therefore, in order to further explore the anti-depressive mechanism of TCM, this paper proposes research strategies on the anti-depressive mechanism of TCM based on functional metabolomics from the perspective of metabolism, the potential biomarkers of depression are analyzed with the help of multi-omics combined analysis technology, and the functional molecules of TCM for antidepressant are studied. Molecular biology techniques are used to accurately capture the molecular interactions between biomarkers of depression and functional compounds, which identify effective drug targets and further elucidate the biochemical functions and related mechanisms involved in depression metabolic disorders. This paper systematically reviews the research strategies and applications of functional metabolomics in the anti-depressive mechanisms of TCM, expounds on the core value of functional metabolomics, and summarizes the current research status and hot issues of TCM for antidepressants in recent years, providing new methods and new ideas for the study of mechanisms of TCM with the help of functional metabolomics.

ABSTRACT

Objective To establish Metrigel-VEGF-SW982 complexes and use the complexes to produce animal models of synovial sarcoma so as to provide new ideas for establishing other models of soft tissue tumors. Methods After the SW982 cells were cultured and collected,they were resuspended with Metrigel,and VEGF was added.The suspension was seeded into transwell to establish the scaffold complexes of Metrigel-VEGF-SW982.The complexes were cultured overnight.Cryosections were made and HE staining was carried out to observe the cell scaffold complexes.We randomly divided 10 female SCID mice(4 week old)into scaffold group and control group. The mice in the scaffold group were transplanted with cell-scaffold complexes,and the control group with cell suspension.After 8 weeks,the success rate of modeling was compared between two groups.The mice were sacrificed and the tumors were obtained.HE staining was carried out to observe the histopathological features of tumors in both scaffold group and control group.Results The SW982 cells were cultured with Matrigel in a 3D way,which could simulate the growth condition of cells in vivo.Establishing synovial sarcoma animal model with cell scaffold complex could increase the success rate.The tumors in scaffold group had a larger volume,higher density of tumor cells and greater vascularization(P<0.05).Conclusion Establishing synovial sarcoma animal model with Metrigel-VEGF-SW982 complex can greatly improve the success rate of modeling,which can provide basis for the study of synovial sarcoma.

ABSTRACT

Patients with bilateral vertebral artery occlusion have a high incidence of cerebral infarction with poor prognosis. Infarction of bilateral middle cerebellar peduncle (MCP) is extremely rare and only a few cases have been reported in literature. A 74-year-old male patient was admitted to our hospital with a chief complaint of dizziness and walking instability for 13 d. Brain magnetic resonance image showed acute bilateral middle cerebellar peduncle infarction. Digital subtraction angiography showed occlusion of the initiation part of left vertebral artery and whole right vertebral artery, while a large amount of collateral circulations and recanalization were observed. After volume expansion, anti-platelet aggregation and lipid-lowering therapy, the symptoms disappeared. The patient was followed up for 10 months and he recovered well.

ABSTRACT

Spleen kidney Yang deficiency (SKYD) diarrhea is a common syndrome in tranditional Chinese medicine (TCM). Until now, there is not an ideal SKYD diarrhea rat model for the research. In this study, we compared single factor way (method I, injecting hydrocortisone and gavaging Sennae Folium) with compound factors way(method II, gavaging adenine, improper diet, exhaustion, and gavaging Sennae Folium) on establishing SKYD diarrhea rat model. After modelling, diarrhea index, D-xylose excretory rate, NOS/cGMP signal transduction system, organ index and histopathology examination were used to evaluate the two ways. The results showed that, compared with health group, all the assessment criterias of method I and method II had significant differences (P < 0.01, 0.05). In addition, the index such as diarrhea index, NOS/cGMP signal transduction system, organ index (kidney, testis and thymus) and histopathology examination had significant differences (P < 0.01, 0.05) between method I and method II. In conclusion, the compound factors modelling method better conforms to the symptom of diarrhoea model caused by SKYD. This new modelling method provides a basis for studying on TCM astringents warming and tonifying the spleen and kidney, relieving diarrhea.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the early cardiovascular complications of patients underwent lung transplantation.</p><p><b>METHODS</b>The clinical records of 73 patients who underwent lung transplantation in Wuxi People's Hospital from September 2002 to September of 2010 were retrospectively analyzed. All patients were transferred to intensive care unit (ICU) after lung transplantation, received invasive monitoring (PICCO), mechanical ventilation, immunosuppressive therapy and measures to prevent ischemic reperfusion injury. The early cardiovascular complications after lung transplantation up to discharge from hospital were observed.</p><p><b>RESULTS</b>The postoperative mortality was 20.5% (15/73) within 30 days after surgery. Five patients died of cardiovascular reasons including 2 cases of pulmonary embolism and 3 cases of ventricular fibrillation. Cardiovascular complications during the early post-operation period included: paroxysmal atrial fibrillation (19 cases, 26.0%) and persist atrial fibrillation (1 case, 1.4%); atrial fibrillation and atrial flutter in 3 cases and persistent atrial flutter in 1 patient; ventricular fibrillation (3 cases, 4.1%); paroxysmal supraventricular tachycardia (3 cases, 4.1%); ventricular tachycardia (2 cases, 2.7%); bundle branch block (8 cases, 11.0%); intraventricular block(4 cases, 5.5%); left ventricular heart failure(4 cases, 5.5%), right heart failure(6 cases, 8.2%); pulmonary embolism (2 cases, 2.7%), deep venous thrombosis (1 case, 1.4%).</p><p><b>CONCLUSIONS</b>Atrial fibrillation is the most common cardiovascular complication post lung transplantation. Pulmonary embolism and ventricular fibrillation are not common but related with high mortality rate post lung transplantation.</p>

ABSTRACT

OBJECTIVE: To develop an HPLC method for simultaneously determining the contents of naringin and neohespiridin in rat plasma, and to investigate the pharmacokinetics and relative bioavailability of self-microemulsifying pellets of daidai flavones in rats. METHODS: The rats were divided into two groups randomly. They were administered orally suspension of daidai flavones effective parts and its self-microemulsifying pellets, which both contained 120 mg · kg-1 of flavonoids. Blood samples were collected at 0.083, 0.167, 0.333, 0.5, 0.667, 1, 2, 4, 6, 8, 10, 12 and 24 h and the concentrations of naringin and neohesperidin in rat plasma were simultaneously determined by HPLC. The pharmacokinetic parameters were calculated by DAS 3.0 pharmacokinetic program. RESULTS: The pharmacokintic parameters of naringin in the self-microemulsifying pellets of daidai flavones and the suspension were as follows: AUC0-24 (17.710 ± 6.588) and( 9.139 ± 1.982) mg · h · L-1, ρmax (4.816 ± 1.329) and (1.572 ± 0.324) mg · L-1, tmax (0.611 ± 0.086) and (1.575 ± 0.324)h, and t1/2 (13.078 ± 6.382) and (11.678 ± 6.919)h, respectively. And the pharmacokintic parameters of neohesperidin were as follow: AUC0-24 (18.094 ± 4.892) and (10.961 ± 2.276)mg · h · L-1, ρmax (5.657 ± 1.391) and (2.096 ± 0.512) mg · L-1, tmax(0.583 ± 0.091) and (0.806 ± 0.222) h, and t1/2 (14.606 ± 7.562) and (10.985 ± 6.963)h, respectively. The relative bioavailabilities of naringin and neohesperidin in the self-microemulsifying pellets of daidai flavones were 193.78% and 165.08%, respectively. CONCLUSION: The bioavailability of daidai flavones is increased by making them into self-microemulsifying pellets, and the biopharmaceutical property of the flavones is improved.

ABSTRACT

Objective To find out the iodine nutritional status of children in Xiamen island, and to provide the scientific basis for iodine supplimentation. Methods On March 2010, thyroids of all children aged 6to 12, from primary school on the Xiaodeng island of Xiamen were examined by palpation, urinary iodine, iodine content of salt athome and IQ level were tested, and were collected 20 households, iodine content of drinking water was tested randomly. Results IQ testing and thyroid palpation were carried out among a total of 156 children, the goiter rate of children was 1.28% (2/156), the mean IQ was 110; 154 urine samples were taken, the median urinary iodine was 219.1 μg/L; a total of 153 salt samples were tested, and the qualified rate of iodized salt was 87.58%(134/153), and the mean iodine content in the tap water was 4.52 μg/L Conclusions Iodine nutritional status of the island residents is better, and there are no such problems as excessive iodine.

ABSTRACT

Objective To assess the iodine nutritional status of special target population in coastal saltproducing areas and coastal non-salt-producing areas in Xiamen city,and to provide a basis for take appropriate measures for prevention of iodine deficiency disorders.Methods The Xiang-An salt-producing areas and the JiMei non-salt-producing areas were chosen as research spots in 2009.One sample of produced water and 2 samples of tap water were collected to test iodine level; 600 children aged 8 to 10 were selected and thyroid palpation was performed,besides,the urine sample and household salt sample were also collected for iodine determination.Sixty pregnant women,breasffeeding women,and 0 - 2 year old infants were recruited,respectively,and urine samples and household salt samples were collected to perform the determination of iodine level.Results The iodine levels in drinking water of Xiang-An district and Ji-Mei district were 3.23 and 6.05 mg/L,respectively.The consumption rates of edible qualified iodinated salt were 84.4% (438/519) and 98.3% (392/399),respectively.The goiter rates of children aged 8 - 10 were 3.03%(19/628) and 0.67%(4/600),respectively.The medians of urinary iodine were 202.80 and 238.40 μg/L,respectively.The proportions of urinary iodine level < 50 μg/L were 3.5% (14/405) and 1.0%(2/202),respectively.The medians of urinary iodine of the pregnant women were 120.55 and 153.35 μg/L,respectively,and the proportions of urinary iodine level < 150 μg/L were 62.1% (46/74) and 46.8% (29/62),respectively.The medians of urinary iodine in three trimester were 173.10,144.75 and 101.90 μg/L,respectively,early trimester of pregnancy > second trimester and third trimester (Z =6.151,3.052,all P < 0.05),second trimester > third trimester (Z =2.016,P < 0.05 ).The medians of urinary iodine of the breastfeeding women were 131.20 and 104.35 μg/L,respectively.The proportions of urinary iodine level < 100 μg/L were 35.3% (24/68) and 46.7%(28/60),respectively.The medians of urinary iodine of the infants were 81.95 and 80.20 μg/L,respectively,the proportions of urinary iodine level < 100 μg/L were 59.7%(37/62) and 61.6%(40/65),respectively,< 50 μg/L were 32.3% (20/62) and 30.8% (20/65),respectively.Conclusions The levels of iodine nutrition in pregnant women,breastfeeding women,and 0 - 2 year old infants from Xiang-An district and Ji-Mei district in Xiamen city are still below the desired level of iodine nutrition,and the infants and pregnant women in coastal salt-producing areas are poor in iodine nutrition,we should pay close attention.We should strengthen market supervision on iodized salt,carried out iodine nutrition monitoring on pregnant women,breasffeeding women,and infants,and disseminate knowledge of iodine nutrition among high-risk population should be carried out immediately.

ABSTRACT

Objective To investigate the current conditions of water iodine,childrens'iodine nutrition and residents'edible circumstance of iodized salt in the villages with high iodine in drinking water and the adjacent three villages in Xiamen city of Fujian province.Methods Four natural villages of Qianbian,Donglian,Dazhong and Dongshan of Xiangan county were chosen as survey spots in 2008.In each village,one running water sample and all well water samples were collected to obtain the benchmark for each location.All children aged 7-13 year in the four villages underwent thyroid palpation and were selected to measure the urine iodine and the salt iodine.The water iodine and urine iodine were determined by As3+-Ce4+ catalytic spectrophotometry digestion,salt iodine was determined by direct titration.Results In four investigated villages,iodine of four running water samples were all 1.5μg/L.The range of 237 well water iodine samples was from 0.1 μg/L to 506.0 μg/L.There were 18.6% (44/237)specimens in which the well water iodine less than 10 μg/L,73.4%(174/237)between 10 μg/L and 150 μg/L and 8%(19/237)more than 150μg/L.The median of urinary iodine was 153.3 μg/L in 79 urine samples,which was 114%(9/79)more than 200μg/L and less than 300μg/L,12.7%(10/79)equal or more than 300 μg/L and less than 500μg/L,7.6%(6/79)equal or more than 500μg/L and less than 800 μg/L in all samples.Seventy-nine students were examined by palpation and the total goiter rate of children measured was 11.4% (9/79).Seventy-one samples of iodine salt were detected and the coverage rate of iodized salt was 77.5%(55/71).Conclusions The well water iodine contents have a wide distribution in the investigated villages.We should enhance the community awareness by educating them on the damage of iodine excess and iodine deficiency.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of docosahexaenoic acid (DHA) on action potential (AP) and transient outward potassium current (I(to)) on ventricular myocytes of Sprague-Dawley rat.</p><p><b>METHODS</b>Calcium-tolerant ventricular myocytes were isolated by enzyme digestion. The changes of AP and I(to) with increasing DHA at concentrations of 0, 10, 20, 40, 60, 80, 100, 120 and 200 micromol/L were recorded by whole-cell patch clamp configuration.</p><p><b>RESULTS</b>(1) Action potential durations (APDs) were not affected by DHA at concentrations from 0 micromol/L to 30 micromol/L, while APDs were gradually prolonged in proportion with increasing DHA concentrations from 30 micromol/L to 200 micromol/L within 5 minutes and remained stable thereafter. APD(25), APD(50) and APD(75) were (7.7 +/- 2.0) ms, (21.2 +/- 3.5) ms, and (100.1 +/- 9.8) ms respectively at 100 micromol/L DHA. APD(25), APD(50), and APD(75) were (15.2 +/- 4.0) ms, (45.7 +/- 6.8) ms, and (215.6 +/- 15.7) ms respectively at 200 micromol/L DHA. (2) I(to) was gradually reduced with the increasing DHA concentrations from 10 micromol/L to 200 micromol/L. I(to) was blocked by DHA in a dose-dependent manner. I(to) current density was (30.1 +/- 7.2) pA/pF at DHA concentration of 60 micromol/L and its half-inhibition concentration was 58.3 micromol/L.</p><p><b>CONCLUSION</b>APDs are gradually prolonged while I(to) reduced with increasing concentrations of DHA which might contribute to the anti-arrhythmia mechanisms of DHA.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study change and the value of diagnosis of harmonic imaging ultrasonic integrated backscatter (IBS) on the very early diagnosis of acute myocardial infarction (AMI).</p><p><b>METHOD</b>72 patients were divided 2 groups, A group 30 cases with AMI at the very early stage of acute myocardial infarction (in 2 hours), B group 42 cases with typical acute myocardial infarction (in 2 - 12 hours) were examined by IBS and the cyclic variation of integrated backscatter (CVIB) with HP-5500 ultrasonic system in the segment of myocardial infarction and no myocardial infarction.</p><p><b>RESULTS</b>In A group the IBS of the segment of AMI are very higher than that of the segment of no AMI [(18.8 +/- 3.4) dB vs (8.3 +/- 1.2) dB, P < 0.01], the CVIB are lower [(6.3 +/- 0.7) dB vs (7.6 +/- 1.1) dB, P < 0.01]. But in the meanwhile there are no obvious changes in ECG. In B group the IBS of the segment of AMI is obvious higher than the normal person and no infarction segment in the same heart [(22.2 +/- 4.1) dB vs (8.3 +/- 1.2) dB, (21.1 +/- 3.2) dB vs (8.7 +/- 0.9) dB, P < 0.05], but CVIB is obvious lower than the normal person and no infarction segment in the same heart [(5.6 +/- 0.8) dB vs (7.6 +/- 1.1) dB, P < 0.05; (5.8 +/- 0.7) dB vs (9.3 +/- 0.9) dB, P < 0.01], the changes of the ultrasonic is coincidence with ECG. The change of IBS are very obvious just as in ECG.</p><p><b>CONCLUSION</b>The result demonstrate that ultrasonic tissue characterization with harmonic imaging integrated backscatter can be used for diagnosis the very early stage of AMI, and can judge the segment range of AMI and function of the whole heart.</p>

ABSTRACT

<p><b>BACKGROUND</b>The clinical outcome of percutaneous coronary intervention (PCI) is poorer in women than that in men. This study aimed at comparing the impact of gender difference on the strategy of primary PCI in patients with acute ST-segment elevation myocardial infarction (STEMI).</p><p><b>METHODS</b>Two hundred and fifty-nine patients with STEMI who underwent primary PCI within 12 hours of symptom onset were enrolled. The male group consisted of 143 men aged > 55 years, and a female group included 116 women without age limitation. Procedural success was defined as residual stenosis < 20% with thrombolysis in myocardial infarction flow grade > 2 and without death, emergency bypass surgery or disabling cerebral events during the hospitalization. The rate of major adverse cardiac events (MACE), including death, nonfatal myocardial infarction and target vessel revascularization during follow-up, was recorded.</p><p><b>RESULTS</b>Female patients were more hypertensive and diabetic and with fewer cigarette smokers than male counterparts. The prevalence of angiographic 3-vessel disease was higher in the female group, but the procedural success rate was comparable between the two groups (94.4% vs 92.2%). The occurrence rate of MACE did not differ during the hospitalization (4.2% vs 6.0%, P = 0.50), but was significantly higher in the female group during follow-up (mean (16.0 +/- 11.2) months) than that in the male group (5.4% vs 0.7%, P = 0.02).</p><p><b>CONCLUSION</b>Despite a similar success rate of primary PCI and in-hospital outcomes in both genders, female patients with acute STEMI still have a worse prognosis during the long-term follow-up.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the differences of total flavonoid (TF) content and antifree radical activity between the-leaves of bamboo and Gingo biloba, as well as their seasonal changes.</p><p><b>METHOD</b>Spectrophotometery and Chemiluminescence methods were adopted to determine TF and half inhibiting concentration (IC50) on active oxygen free radicals of the leaves of bamboo, phyllostachys nigra (Lodd. ex. Lindl.) Munro, and Ginkgo biloba. Two kinds of leaves were picked in the same plot at the same time monthly.</p><p><b>RESULT</b>The TF of bamboo leaf varied in the range of 0.67%-1.71% (in dry basis of leaf, below as same) throughout a year, the minimum apparing in June and the maximum in July, then going down obviously, and remaining at a much high lever during November to next April. However, the TF of Ginkgo bilabo leaf varied in 1.48%-2.49% during whole growing period, early April to late November. It ascended with the growth of leaf, reaching the top during June and July, the going down slowly, and finally another peak appeared before defoliation. The average IC50 values on O2-. and .OH of bamboo leaf were at 11.0 micrograms.mL-1 and 5.3 mg.mL-1, and Ginkgo biloba at 19.0 micrograms.mL-1 and 3.6 mg.mL-1, respectively.</p><p><b>CONCLUSION</b>The TF content and anti-free radical activity the bamboo leaf are comparable with the leaf of ginkgo biloba, which is a kind of potential resources for natural antioxidant and free radical scavenger.</p>
